Tirzepatide is a synthetic dual agonist peptide that activates both the glucose-dependent insulinotropic polypeptide (GIP) receptor and the glucagon-like peptide-1 (GLP-1) receptor, identified in the literature under development code LY3298176. It was among the first compounds in its class to combine these two incretin targets in a single molecule.
The published evidence base is comparatively strong for a research peptide: multiple phase 3 randomized controlled trials address glycemic control in type 2 diabetes and weight reduction in obesity, with research spanning 2021 through 2025. Head-to-head comparison data against semaglutide has been generated, though long-term cardiovascular outcomes and durability of effect after discontinuation remain open questions.
