Cagrilintide

Cagrilintide is a long-acting, lipidated analogue of amylin, a pancreatic hormone co-secreted with insulin that signals satiety. Natural amylin has a short half-life and tendency to form amyloid fibrils; Novo Nordisk engineered a stabilised, fatty-acid-conjugated molecule capable of once-weekly subcutaneous administration for chronic weight management research.

The compound has accumulated substantial human trial data, including a 26-week phase 2 monotherapy trial and phase 3a combination studies with semaglutide (CagriSema). Independent monotherapy evidence remains limited to the single phase 2 trial, and long-term cardiovascular outcome data has not yet been published.
